On my 08 Bandit, will I need to relocate the rear turn signals if I put on Givi E21 bags?

Not sure about an 08', but on my 2000 with a Givi Wingrack the stock ones had to be removed and wired up to some Fugly Givi ones.

I can't vouch for your bike, but I have a CBR1100XX. I got the Wingrack II with turn signals. They were functional, but ugly. Someone showed me how to "chop" down the stalks on my OEM turn signals which let me mount them within the space left after the Wingrack was installed, and it looked much better.
Perhaps you can do the same or find similar turn signals with shorter stalks?

Zaphod did not want to tangle with them and, deciding that just as discretion was the better part of valor, so was cowardice the better part of discretion, he valiantly hid himself in a closet.

You most likely will have to relocate. I have the tubular side-racks and they provide a spot to re-mount the stock indicators. I never had to do that however since I integrated my indicators in the housing where the brake light is.

I just looked on TwistedThrottle.com and they show the stock indicators relocated w/ the tubular racks which would hold the E21 bags:
